Decreased expression of E3 ubiquitin ligases during ischemia–reperfusion. Isolated rat hearts were perfused under high glucose conditions (33 mM glucose) vs. controls (11 mM glucose) for 60 min and subsequently subjected to 20 min of global ischemia, followed by 60 min of reperfusion ± either 5 μM lactacystin or 10 μM MG-132 treatment for the first 20 min of reperfusion. Western blot analysis was completed for: a muscle RING finger 1 (MuRF1) and b muscle atrophy F-box (MAFbx) with lactacystin treatment; c MURF1 and d MAFbx with MG-132 administration. We employed β-actin as loading control. Values are expressed as mean ± SEM (n ≥ 4). *p < 0.05, **p < 0.01 vs. respective controls.
