Glibenclamide inhibited IL-1β expression by high glucose and LPS stimulation in primary peritoneal macrophages. IL-1β and TNF-α in the supernatants of macrophages were assessed by ELISA. Macrophages were incubated for 24 h with high glucose (33 mM), or high glucose + LPS (1 μg/ml), or high glucose + LPS + glibenclamide (100 μM). Cells incubated without drug treatment served as control. Either high glucose or High glucose + LPS induced significant increase of both IL-1β and TNF-α, glibenclamide treatment only significantly inhibited IL-1β expression by high glucose + LPS stimulation. *P < 0.05, **P < 0.01 vs control; ##P < 0.01 vs Glucose + LPS, †P < 0.05 vs LPS.
