Representative cardiomyocyte sarcomere shortening-relengthening transients with (−−-) and without (—) 32 μ M extracellular Zn2+and intracellular Ca2+transients. A and B. At 2 Hz cardiomyocyte shortening was greater in HG compared to nonHG. Diastolic sarcomere length was noticeably shorter in HG compared to nonHG at 2 Hz. Extracellular Zn2+ reduced cardiomyocyte shortening in both HG and nonHG and led to longer diastolic sarcomere lengths in HG. C and D. At 6 Hz extracellular Zn2+ reduced cardiomyocyte shortening in both HG and nonHG. Diastolic sarcomere length was still noticeably shorter in HG compared to nonHG, and exposure to Zn2+ led to longer diastolic sarcomere lengths in the HG group. E and F. At 2 and 6 Hz, fluorescence ratio transients of HG and nonHG cardiomyocytes were similar in temporal characteristics, but there was a greater area under Fura-2 fluorescence ratio transient in the HG at 2 Hz and a trend at 6 Hz.
